A pathomorphological definition of a hydatid cyst located on the facies diaphragmatica of the spleen in a 1.5 year old sheep provided from a slaughterhouse was performed in this study.
Case Report
The spleen belonging to a 1.5 year old sheep revealed to have a hydatid cyst. The material was processed in accordance with routine histological procedures, fixed in 10% formalin solution and stained with Hematoxylin and Eosin (HE).Macroscopical examination revealed the presence of a single cyst filled with clear fluid located in the center of the facies diaphragmatica of the spleen, detected to protrude from the surface and to measure 3.5x3.5x4 cm (Fig 1). The cyst was observed to be distinctively separated from surrounding tissue with a thin and light yellow colored wall. Upon drainage of the fluid, the inner surface of the cyst wall was observed to gain a folded appearance and to display numerous white colored granular structures (Fig 2).

On the other hand, microscopical examination revealed two different appearances specific to the cyst and the host. The outer wall of the cyst was detected to be light pinkish in color and to be formed of hyalinous cuticular membrane (Fig 3).

The germinative layer was assessed to exist below the outer wall. Necrotic material and many giant cells, surrounded by an outer layer comprised of macrophages, lymphocytes and a few numbers of epithelioid histiocytes were present at the periphery of the cyst wall (Fig 4). The area in which the cyst was located was surrounded by a thin fibrous capsule from the external.

Discussion
Hydatidosis (echinococcosis) caused by Echinococcus granulosus and Echinococcus multilocularis is a parasitic infection related closely both to human and animal health and known to cause economic loss. 1,2 The adult parasite develops in carnivores whereas larvae develop in primarily sheep and cattle especially in omnivores, herbivore animals and humans. 3,4,5,6 Hydatid cysts are encountered in cattle and sheep at rates of 7.8-50% and 3-11.4%, respectively. 2,7 These cysts which may be one or more in number, contain a clear fluid and are most frequently located in the lungs and liver as well as other internal organs including the spleen, kidneys and brain. 2,3,4,7,8,9,10 Cysts may be present in one or more of the above mentioned internal organs. In this reported case, the hydatid cyst was located in the spleen, an internal organ in which hydatidosis is less frequently observed. Macroscopical and microscopical findings of the cyst were determined to be in accordance with literature. 3,8,9,11 However, although some researchers 12,13 have reported the thickness of the cyst capsule which develops due to fibrous host reaction to be dependent upon the severity of the reaction and the size of the cyst to be related to the features of the tissue the cyst is located in, we believe that such a generalization would be misleading for internal organs in which hydatidosis is less frequently observed, including the spleen. However the present study depicts the cloning and characterization of the cDNA sequence of 497bp encoding conglutinin (neck and carbohydrate recognition domain) from sheep for the first time and edited sequence of 488bp was submitted to NCBI with accession number JQ692170. Moreover sequence information revealed that nucleotide sequence of sheep conglutinin was 93.4% homology to the bovine conglutinin, 93.9% to buffalo conglutinin, 93.4% to nilgai conglutinin, 81.6% to CL-43 and 90.2% to CL-46 also liver is known to be the organ for the presence of this novel gene in this small ruminant for first time. Oral administration of troleandomycin at a dose of 100 mg/kg/day for 6 days to three adult male Lacaune sheep produced a 1.6-fold increase in specific content of liver microsomal cytochrome P-450. In sodium dodecyl sulfate-polyacrylamide gel electrophoresis, microsomal preparations from treated animals exhibited a strong band in the zone of electrophoretic mobility of cytochromes P-450. This band corresponded to a cytochrome P-450 which cross-reacted with rabbit P450IIIA6 antibodies, as demonstrated by immunoblotting. The ovine isozyme was purified to electrophoretic homogeneity by means of successive DEAE cellulose, CM cellulose and hydroxylapatite chromatographic separations. This hemoprotein had an apparent molecular weight of 52 kD as determined by calibrated sodium dodecyl sulfate-polyacrylamide gel electrophoresis and was characterized in terms of spectral data, NH2-terminal amino acid sequence, immunologic and catalytic properties. This study revealed some interspecies differences with the orthologous rabbit isozyme. The contribution of this form to the N-demethylation of erythromycin and of three veterinary drugs: chlorpromazine, chlorpheniramine and bromhexine was demonstrated from inhibition by TAO, from immunoinhibition studies, using polyclonal antibodies raised in rabbit and from the existence of significant correlations between its microsomal level and these N-demethylase activities. In contrast, the results suggest that ovine P450IIIA could not be predominantly involved in the N-dealkylation of benzphetamine, ephedrine, ivermectine or spiramycin.

5.47AbstractComparative histology deals with the comparison of microscopic structural relations of the various animals with in the ecosystem. Here, we compare the microscopic structure of the human liver with domestic animals like cow, sheep and goat. Human and cow, goat and sheep's liver were taken and divided in to 3 groups. We kept liver specimen in formalin for fixation. Thin cut sections of specimen were taken after paraffin embedding. Slides were stained by Haematoxylene and Eosin, later observed the histological features under light microscope. The study was undertaken to compare the histological differences like hepatic lobule, connective tissue septa, portal triad, hepatocytes of liver between human and cow, goat and sheep. It plays a useful tool for morphological studies based on the evolution. Hepatic lobule was hexagonal in shape in cow, goat and sheep, but it was not clearly seen in human liver. Hepatocytes were larger in human beings but smaller and polygonal in cow, goat and sheep. Connective tissue septa were scanty in human liver, in comparison to other animals. Central vein was closer to the hepatic lobule in human and goat's liver, while in case of cow and sheep, it was found to be close to the portal triad. This comparative histological study may be useful to all the research scholars who undertaken similar studies, veterinary scientists and the field of liver transplantation.Do you want to read the rest of this article?Request full-text
