济南大学硕十学位论文 摘 要 随着免疫组织化学技术的不断发展，免疫组化已经成为当今临床病理诊断和研究
的重要辅助手段。计算机图像处理技术以其精确性、客观性、处理速度快等方面的优
势，已经成为免疫组化显微图像分析技术的发展趋势。但是，目前很多学者在免疫组
化图像自动分割方面的研究成果主要局限在对一些特征明显的组织结构进行自动分
割，对免疫组化图像的分析方法也主要是基于单个参数和特定图像，这些方法都有其
局限性，导致最终分析结果不够全面、客观、准确。因此，研究免疫组化显微图像相
关的分割和分析技术，辅助医生准确观察和定量检测免疫组化显色反应强度，已经成
为国内外学者当前和今后研究的热点。 本课题以卵巢癌免疫组化显微图像为主要研究对象，重点研究了阳性细胞区域的
分割技术和免疫组化阳性表达结果的分析技术，并且在分割和分析方法上都有所创
新。论文的研究工作主要包括以下几个方面： 1 研究了免疫组化显微图像的相关病理学知识。包括免疫组化染色的方法、免
疫组化图像的显微特征和免疫组化图像的分类等信息，为后续正确的分割和分析技术
的研究奠定基础。 2 研究了图像预处理和颜色空间转换的相关理论和方法。在图像增强方法和颜
色空间的选择上都做了大量的对比实验，最后选择比较适合免疫组化显微图像的图像
增强方法和适合其分割的颜色空间。 3 研究了免疫组化显微图像的分割技术。主要采用了如下三种方法：方法l是
基于色度学准则的ISODATA聚类分割方法。在RGB颜色空间下，利用色度学准则
和ISODATA聚类方法分割提取阳性细胞区域。方法2是基于ⅥⅣ颜色空间的OTSU
